The native hormone is produced in the hypothalamus and acts on the anterior pituitary. Binding of GHRH to its receptor stimulates synthesis and release of growth hormone into circulation. Because the analogue retains the receptor-binding region of the parent sequence, it engages the same receptor and triggers the same downstream signaling. The result is increased growth hormone secretion from pituitary cells, which in turn influences hepatic production of insulin-like growth factor 1. This axis is the basis for the compound's measured biological effects.
Interest in this peptide developed because native GHRH has a short circulating lifetime. The N-terminal modification slows cleavage by dipeptidyl peptidase IV, an enzyme that removes the first two residues of many peptides and terminates their activity. Slower degradation means a longer window of receptor stimulation per administration. This design logic parallels other modified peptide hormones, where a small chemical change at a vulnerable site yields a more durable molecule without altering the core mechanism of action.
The peptide is synthesized chemically rather than extracted from biological sources. Solid-phase synthesis builds the chain from the C-terminus toward the N-terminus, after which the hexenoyl group is attached. Purity is typically assessed by high-performance liquid chromatography, and identity is confirmed by mass spectrometry. Regulatory review of the finished product focuses on these analytical controls, since small deviations in sequence or modification can change biological activity. Questions about long-term effects on the pituitary axis remain areas of continued investigation.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone, composed of 44 amino acids. It was designed to retain the biological activity of the native hormone while resisting rapid enzymatic degradation. The compound is classified as a growth hormone secretagogue and belongs to the broader family of hypothalamic releasing factors. In research and clinical settings, it is studied for its ability to stimulate pituitary growth hormone release. Its structure includes a modification at the N-terminus that contributes to an extended half-life relative to native growth hormone-releasing hormone.
Tesamorelin binds to growth hormone-releasing hormone receptors on the surface of pituitary somatotroph cells. This binding activates adenylate cyclase, raising intracellular cyclic AMP levels and triggering the release of growth hormone into circulation. The elevated growth hormone then stimulates hepatic production of insulin-like growth factor 1. Because the effect is mediated through the endogenous axis, secretion remains subject to feedback regulation. This distinguishes it from direct growth hormone administration, which bypasses pituitary control entirely.

It appears in the sex cord-stromal tumour group of ovarian neoplasms. Ovary fibromas are most frequent during middle age, and rare in children. Upon gross pathological inspection, ovary fibromas are firm and white or tan. Variants with edema are especially likely to be associated with Meigs' syndrome. On microscopic examination, there are intersecting bundles of spindle cells producing collagen. There may be thecomatous areas (fibrothecoma). The presence of an ovarian fibroma can cause ovarian torsion in some cases.
In sensory neurons, an external signal such as pressure, temperature, light, or sound is coupled with the opening and closing of ion channels, which in turn alter the ionic permeabilities of the membrane and its voltage. These voltage changes can again be excitatory (depolarizing) or inhibitory (hyperpolarizing) and, in some sensory neurons, their combined effects can depolarize the axon hillock enough to provoke action potentials. Some examples in humans include the olfactory receptor neuron and Meissner's corpuscle, which are critical for the sense of smell and touch, respectively. However, not all sensory neurons convert their external signals into action potentials; some do not even have an axon. Instead, they may convert the signal into the release of a neurotransmitter, or into continuous graded potentials, either of which may stimulate subsequent neuron(s) into firing an action potential. For illustration, in the human ear, hair cells convert the incoming sound into the opening and closing of mechanically gated ion channels, which may cause neurotransmitter molecules to be released. In similar manner, in the human retina, the initial photoreceptor cells and the next layer of cells (comprising bipolar cells and horizontal cells) do not produce action potentials; only some amacrine cells and the third layer, the ganglion cells, produce action potentials, which then travel up the optic nerve.
There were several terrorist attacks in Xinjiang in 2013 and 2014; an attack in Ürümqi in April 2014 occurred just after the conclusion of a visit by Xi to Xinjiang. Following these attacks, CCP leaders held a secret meeting to find a solution, leading to Xi to launch the Strike Hard Campaign Against Violent Terrorism in 2014, which involved mass detention, and surveillance of ethnic Uyghurs there. The campaign included the detainment of 1.8 million people in internment camps, mostly Uyghurs but also including other ethnic and religious minorities, by 2020, and a birth suppression campaign that led to a large drop in the Uyghur birth rate by 2019. Human rights groups and former inmates have said Uyghurs and other minorities have been forcibly assimilated into China's majority ethnic Han society in the camps. This program has been called a genocide by some observers, while a report by the UN Human Rights Office said they may amount to crimes against humanity.

=== Derivatives === Various alginate-based materials can be produced, including porous scaffold material, alginate hydrogel, nonwoven fabric, and alginate membranes. Techniques used to produce these include ion cross-linking, microfluidic spinning, freeze drying, wet spinning, and immersive centrifugal jet spinning. Calcium salts added to a sodium alginate solution to induce ionic cross-linking, which produces the hydrogel. Freeze-drying the hydrogel to eliminate water produces the porous scaffold material. Wet spinning consists of extruding an alginate solution from a spinneret into a calcium salt solution to induce ionic cross-linking (forming the gel), and then drawing the fibers out of the bath with draft rollers. Microfluidic spinning, a simpler and more eco-friendly implementation of the process, involves introducing calcium salt flows flowing alongside and touching a central "core" flow of alginate. These flows form a "sheath". The fiber then emerges from the core flow. This technique can be used to produce shaped and grooved fibers.
The rate of degradation is dependent on the type of injury and is also slower in the CNS than in the PNS. Another factor that affects degradation rate is the diameter of the axon: larger axons require a longer time for the cytoskeleton to degrade and thus take a longer time to degenerate.
=== Petra Koenig === Petra Koenig (Sarah Goldberg) is a senior portfolio manager at FutureDawn Partners and one of Anna Gearing’s closest associates. Despite their long professional proximity, Petra’s relationship with Anna has grown strained over time. Although Petra is godmother to Anna’s children, she later reflects that the role was imposed by Anna’s seniority rather than freely chosen, and that Anna pushed for a personal closeness Petra did not seek. Petra’s pragmatic, results-driven approach to investing increasingly clashes with Anna’s rigid commitment to FutureDawn’s socially conscious ethos, which Petra regards as performative and strategically naïve. In series 3, Petra becomes disillusioned with Anna’s leadership after Harper—then working at FutureDawn—helps her hedge exposure during the chaotic Lumi IPO by arranging credit default swaps on oil and gas assets through data she obtains from Yasmin. Impressed by Harper and frustrated by Anna’s constraints, Petra leaves FutureDawn to cofound a hedge fund with Harper, which they name LeviathanAlpha. The pair secure seed funding from Otto Mostyn and retain Pierpoint as their broker, before deciding to short Pierpoint after learning the firm’s ESG-backed debt is nearing maturity and effectively unsellable. Their plan unravels when Harper admits she first learned of the debt crisis through an improper disclosure, rendering the trade illegal. Petra alerts Otto, then reconciles with Harper to unwind the short and sever ties with him.

It shares the 44-residue sequence of human GHRH but carries an added trans-3-hexenoyl group at its N-terminus. That addition does not occur in the natural hormone and serves mainly to resist enzymatic breakdown. The receptor target and signaling pathway remain the same.
It binds the growth hormone-releasing hormone receptor on anterior pituitary cells. Activation of that receptor promotes synthesis and release of growth hormone. The effect propagates through the growth hormone and insulin-like growth factor 1 axis.
Native GHRH is cleared quickly by peptidases, which limits how long it can stimulate its receptor. The added group hinders one of the primary cleavage enzymes. The practical consequence is a longer period of receptor activity per dose.
It is a synthetic analog of growth hormone-releasing hormone, a hypothalamic peptide. It functions as a growth hormone secretagogue acting at pituitary receptors. The classification separates it from direct growth hormone products.
