A documented effect of tesamorelin is a reduction in visceral adipose tissue in some study populations. Researchers have reported decreases in trunk fat measured by computed tomography alongside changes in lipid markers. The mechanism is thought to involve growth hormone-mediated lipolysis, though the precise contribution of direct versus indirect pathways is not fully resolved. Studies have generally examined defined groups over finite periods, so long-term outcomes are less well characterized. Findings have not been uniform across all trials.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone (GHRH). Its sequence corresponds to the 44-amino-acid form of human GHRH with a trans-3-hexenoyl group attached to the N-terminal tyrosine. This modification slows enzymatic cleavage and extends the peptide's activity relative to the native hormone. The compound is produced by solid-phase peptide synthesis and supplied as a lyophilized powder. Researchers classify it as a GHRH receptor agonist. Its structure places it in the same family as other growth hormone secretagogues that act on the pituitary.
Measured responses usually involve growth hormone and insulin-like growth factor 1, known as IGF-1. Growth hormone rises in bursts and is difficult to sample reliably, while IGF-1 shifts more slowly and can be assessed from a single blood draw. Studies therefore treat IGF-1 as the more practical pharmacodynamic marker. Both are indirect, showing that the receptor was engaged rather than that the peptide reached a particular concentration. Direct exposure measurement requires an assay aimed at the molecule itself.
Published work tends to frame tesamorelin as a tool for studying the GHRH axis and as a compound with measurable effects on body composition. Reports often describe visceral adipose tissue as an endpoint, assessed by imaging rather than by inference. Analytical sections commonly describe liquid chromatography with tandem mass spectrometry to confirm identity and purity, because immunoassays may cross-react with related fragments. Where results diverge between studies, differences in assay choice, sampling timing, and population are frequent explanations offered. Whether effects persist after treatment stops remains an open question.
Tesamorelin binds the growth hormone–releasing hormone receptor on pituitary somatotroph cells. The receptor signals through the Gs protein, raising intracellular cAMP and activating protein kinase A. That cascade triggers release of stored growth hormone in pulses rather than a steady stream. Because the drug acts at the receptor that normally controls this process, its effect depends on the body's own signaling architecture rather than on a synthetic pathway. The resulting hormone profile reflects the timing of each pulse, not only its size.

Clinical investigation has focused on HIV-associated lipodystrophy, a condition in which antiretroviral therapy contributes to abnormal fat distribution. Excess visceral adipose tissue accumulates in the abdomen while peripheral fat may be lost. Tesamorelin was evaluated for reducing this visceral fat depot, with trials measuring changes in abdominal fat by imaging rather than by body weight alone. The rationale rests on the known lipolytic effects of growth hormone. Effects on visceral fat are documented, while long-term outcomes regarding cardiovascular risk remain less clearly established.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone, composed of 44 amino acids. It was designed to retain the biological activity of the native hormone while resisting rapid enzymatic degradation. The compound is classified as a growth hormone secretagogue and belongs to the broader family of hypothalamic releasing factors. In research and clinical settings, it is studied for its ability to stimulate pituitary growth hormone release. Its structure includes a modification at the N-terminus that contributes to an extended half-life relative to native growth hormone-releasing hormone.
Pharmacodynamic studies show that tesamorelin reduces visceral adipose tissue more than subcutaneous adipose tissue in the studied population. This selectivity may relate to differences in blood flow and hormone sensitivity between fat depots. Effects on glucose metabolism and insulin sensitivity have been investigated, with some trials reporting modest changes and others showing stability. The precise relationship between growth hormone exposure, IGF-1 levels, and visceral fat loss remains an active area of analysis.
Tesamorelin binds to growth hormone-releasing hormone receptors on somatotroph cells in the anterior pituitary. Receptor activation increases intracellular cyclic AMP and promotes synthesis and secretion of growth hormone. Because the peptide mimics endogenous GHRH, it amplifies the normal pulsatile release of growth hormone rather than providing exogenous growth hormone directly. This upstream action distinguishes tesamorelin from recombinant growth hormone preparations and from growth hormone secretagogues that act at different receptors.
== Contraindications == Levothyroxine is contraindicated in people with hypersensitivity to levothyroxine sodium or any component of the formulation, people with acute myocardial infarction, and people with thyrotoxicosis of any etiology. Levothyroxine is also contraindicated for people with uncorrected adrenal insufficiency, as thyroid hormones may cause an acute adrenal crisis by increasing the metabolic clearance of glucocorticoids. For oral tablets, the inability to swallow capsules is an additional contraindication.
In 2009, the Justice Department reported that Mexican drug cartels distribute drugs in nearly 200 cities across the United States, including Los Angeles, Chicago, and Atlanta. Gang-related activity and violence has increased along the U.S. Southwest border region, as U.S.-based gangs act as enforcers for Mexican drug cartels. In October 2025, Mexican cartels are issuing bounties up to $50,000 for a hit on ICE and CBP agents according to the Department of Homeland Security.
An electron capture detector most often uses a radioactive source to generate electrons used for ionization. Some examples of radioactive isotopes used are 3H, 63Ni, 85Kr, and 90Sr. The gas in the detector chamber is ionized by the radiation particles. Nitrogen, argon and helium are common carrier gases used in the ECD. Argon and helium need to be combined with another gas, such as methane, in order to prevent immediate conversion into metastable ions. The combination will extend the lifetime of the metastable ions (10−6 seconds). The methane will cool the electrons during the collisions. The addition of methane will enhance the ability to form negative ions under high pressure because it will adjust the thermal energy to be similar to the energy distribution of the ions. Methane is the most common gas used because it can produce many positive ions when it collides with electrons. These positive ions will then form low energy electrons used for ionization:
Preservation techniques are needed to prevent fish spoilage and lengthen shelf life. They are designed to inhibit the activity of spoilage bacteria and the metabolic changes that result in the loss of fish quality. Spoilage bacteria are the specific bacteria that produce the unpleasant odours and flavours associated with spoiled fish. Fish normally host many bacteria that are not spoilage bacteria, and most of the bacteria present on spoiled fish played no role in the spoilage. To flourish, bacteria need the right temperature, sufficient water and oxygen, and surroundings that are not too acidic. Preservation techniques work by interrupting one or more of these needs. Preservation techniques can be classified as follows.

Broadcasting is the one-way transmission of information from a transmitter to receivers belonging to a public audience. Since the radio waves become weaker with distance, a broadcasting station can only be received within a limited distance of its transmitter. Systems that broadcast from satellites can generally be received over an entire country or continent. Older terrestrial radio and television are paid for by commercial advertising or governments. In subscription systems like satellite television and satellite radio the customer pays a monthly fee. In these systems, the radio signal is encrypted and can only be decrypted by the receiver, which is controlled by the company and can be deactivated if the customer does not pay. Broadcasting uses several parts of the radio spectrum, depending on the type of signals transmitted and the desired target audience. Longwave and medium wave signals can give reliable coverage of areas several hundred kilometers across, but have a more limited information-carrying capacity and so work best with audio signals (speech and music), and the sound quality can be degraded by radio noise from natural and artificial sources. The shortwave bands have a greater potential range but are more subject to interference by distant stations and varying atmospheric conditions that affect reception. In the very high frequency band, greater than 30 megahertz, the Earth's atmosphere has less of an effect on the range of signals, and line-of-sight propagation becomes the principal mode.
protein folding The physical process by which the linear chains of amino acids (i.e. polypeptides) synthesized during translation are changed from random coils into stable, orderly, three-dimensional shapes (i.e. proteins) by assuming a higher-order structure or conformation which permits the protein to be biologically functional, known as its native state. Folding is the consequence of amino acid residues participating in intermolecular electrostatic interactions with each other and with their surroundings, including other molecules, and so is strongly influenced by the particularities of the local chemical environment. The time it takes to properly fold a protein can vary greatly, but the process often begins while chain synthesis is still ongoing. Some chains may have motifs or domains which lack intrinsic order and remain unfolded across a wide range of chemical conditions. Having the correct three-dimensional structure is essential for proper protein function, and misfolded proteins are generally biologically inactive, though mutant folds can occasionally modify functionality in useful ways.
The magnetic field in a solar active region above a sunspot can store energy that is suddenly released as a burst of motion, X-rays, and radiation when the main current sheet collapses and reconnects the field.
=== 7 July === Russian forces claimed to have taken the settlement of Chyhari (part of Pivdenne) in Donetsk Oblast. Four people were killed after their vehicle struck a Russian-planted mine in Kharkiv Oblast. Russian officials claimed that Iskander ballistic missiles destroyed two Patriot missile launchers in Odesa. The Ukrainian Air Force confirmed the attack but provided no further information. The Ukrainian military claimed to have shot down a Russian Su-25 fighter jet over Donetsk Oblast. Russian officials claimed that falling debris from Ukrainian drones set fire to a warehouse in Voronezh Oblast which contained explosives. Another drone was reported shot down over Belgorod Oblast. Explosions were reported at a number of locations around occupied Melitopol, including the Melitopol Air Base, the railway station and other areas. Locals said that Russian air defence was activated during the incident.
=== Complementary and alternative medicine laboratories === Complementary and alternative medicine tests, such as live blood analysis (LBA), biological terrain assessment (BTA), dental sensitivity testing, and cytotoxic testing have not been categorized by the CDC, and are thus treated as high complexity laboratory developed tests. CAM tests are often ordered by chiropractors, naturopaths, and nutritionists and accompanied by non-validated clinical interpretations which are used to recommend or justify costly, unnecessary, and potentially damaging treatments. A 2001 OIG report found that the majority of laboratories performing CAM tests were not enrolled in CLIA and that CAM laboratory personnel did not meet the high complexity qualifications. For laboratories enrolled in CLIA, they had their certificates revoked or were sent deficiency notices for failing to adhere to CLIA regulations. No CLIA laboratory has been able to validate its LBA CAM tests per standards. CAM tests are not covered by health insurance. The number of CAM laboratories is unknown; the sales of supplemental remedies based on the results of these tests is unknown. Since CLIA does not regulate the clinical validity/usefulness of a test, it is possible for a CLIA laboratory to offer tests that have no clinical utility. Getting laboratories that conduct CAM testing to enroll in CLIA is itself a challenge. Medical laboratories enroll in CLIA to qualify for Medicare/Medicaid reimbursement; nearly all providers bill patients directly for CAM laboratory tests.

It mirrors the 44-residue form of human growth hormone-releasing hormone. A hexenoyl group on the N-terminal tyrosine distinguishes it from the unmodified hormone. The change is intended to improve resistance to enzymatic breakdown.
The N-terminal modification reduces cleavage by circulating peptidases, so the peptide persists longer than native GHRH. That persistence is the main rationale for the synthetic design. Comparative half-life values in humans are reported in regulatory review documents rather than in general reference literature.
Reductions in visceral adipose tissue have been measured in controlled studies of defined populations. Whether the effect generalizes to other groups and persists after treatment stops is less clear. Longer-term outcome data remain limited.
It acts on the growth hormone–releasing hormone receptor, a Gs-coupled receptor found on pituitary somatotroph cells. Activation raises cAMP and prompts pulsatile hormone release.
